Evaluation of the microrheological properties of the blood in patients with type 2 diabetes mellitus, using a developed flow microchamber

Abstract: The aim of the present study is to investigate in vitro microrheological properties of the blood (aggregation and deformability of erythrocytes) in patients with type 2 diabetes mellitus under flow conditions, using a developed flow microchamber system.
A flow microchamber (FMC) system has been developed for the analysis of the rheological properties of blood under physiological conditions, which enables to perform in vitro studies, simulating in vivo conditions of the blood vessels.
Whole blood samples from patients with T2DM (n=15) and healthy donors (n=15) were used to prepare diluted suspensions from erythrocytes in an isotonic buffer solution with Dextran 250. These samples were used to determine the erythrocyte aggregation index (EAI) and erythrocyte elongation index (EEI).
The results show that the average values of EAI in patients with T2DM were increased by 23 %, compared to the results of controls. The average values of EEI in patients with T2DM decreased at all studied shear rates (from 0 s-1 to 610 s-1), compared with the data in healthy individuals. Therefore, while erythrocyte aggregation in patients with T2DM increased, erythrocyte deformability decreased, compared to the control group.
The result obtained with FMC could serve as objective biomarkers and tools for diagnosis, stratification of the disease, monitoring, and prediction of microvascular impairments in T2DM.
