Bonding performance of a newly developed one-step single-bottle adhesive on sound human dentin with different tubule orientations

Abstract: Objective. To investigate the bonding performance of a newly developed one-step universal adhesive in relation to the orientation of dentinal tubules.
Methods. The bonding abilities of the Clearfil Universal Bond Quick (CUBQ) single-step adhesive and Clearfil Megabond 2 (MB2; control) were compared in freshly extracted human third molars. Specimens were prepared and machined to obtain parallel or perpendicular orientations of dentin tubules to the bonded surface. The adhesive systems were applied according to the manufacturer’s instructions, following which composite resin material was applied and polymerized by light curing. Beam-shaped specimens containing dentin, adhesive layer, and composite material were obtained from restored teeth and subjected to tensile loads until complete fracture in order to determine the tensile strength. Morphological characteristics of the adhesive interfaces were evaluated in relation to their mechanical behavior by scanning electron microscopy (SEM).
Results. Specimens with perpendicular tubule orientation in the control and CUBQ groups exhibited micro-tensile bond strength values of 65.6 ± 18.0 and 56.1 ± 18.7 MPa, respectively; the corresponding values in specimens with parallel orientation were 45.8 ± 12.3 and 50.4 ± 13.4 MPa, respectively. Fractographic findings of bonded surfaces in both groups revealed mostly cohesive failure within the adhesive resin, regardless of tubule orientation (type A). In the CUBQ group, none of the specimens with parallel and perpendicular orientations exhibited adhesive (type B) and mixed (type C) failure, respectively.
Clinical significance. The very good adhesive abilities of CUBQ make it a preferred choice where speed and ease of use are of primary importance.
