Alterations in hematological parameters after neonatal or prepubertal induction of diabetes mellitus in rat

Резюме: The purpose of this study was to investigate the alterations in hematological, biochemical parameters and spleen index in adult rats after neonatal or prepubertal induction of diabetes mellitus (DM). To induce DM male pups were i.p. injected with streptozotocin (STZ) with a single dose of 100 mg/kg b.w. on day 1 (neonatally, NDM) or on day 10 (prepubertally, PDM). On day 50 spleens were excised, weighed and organ index was calculated. Whole blood was collected and hematological parameters and fasting glucose were evaluated. Spleen index of PDM rats was considerably affected. Significant glucose elevation and changes in red blood cell (RBC) parameters were observed in adult animals from both experimental groups. Mean RBC volume was significantly reduced as well as the hemoglobin-related parameters – MCH and MCHC. Neonatally but not prepubertally induced diabetes considerably increased platelet count and plateletcrit. NDM caused significant rise of monocyte and granulocyte number and slight insignificant decrease of lymphocyte count. These results support data that inflammation and changes in blood cell activity are involved in the pathophysiology of diabetes. The observed changes in blood parameters suggest alterations in hemorheology, coagulation and microcirculation of the diabetic animals.
