Gasotransmitters (NO, CO and H2S) affect the microrheology of old red blood cells more effectively than of young ones: in vitro study

Резюме: Background: It is known that red blood cells (RBCs) of different ages are present in the bloodstream: "young", "mature" and "old" red blood cells (RBCs). At the same time, old RBCs have lower deformability (RBCD) and increased aggregation (RBCA) in comparison with young and mature ones. Under the influence of one of the gasotransmitters (GTs), nitric oxide (NO), there is a change in the RBC microrheology. However, the data on the effect of GT on RBC microrheology of the different ages are contradictory.
Objective: the aim of this study was a comparative analysis of microrheological responses of RBCs of different ages to the action of three gasotransmitters (NO, CO, and H2S).
Methods: In experiments with RBCs divided in a density gradient into young, old, and mature cells (RBCtop, RBCbot and RBCmid), their deformability (RBCD) and aggregation (RBCA) were recorded after cell incubation with GT donors and stimulators of their synthesis.
Results: It was found that GT donors increased RBCD in all three-cell fractions, but to a greater extent in a subpopulation of older RBCs. A similar pattern of changes was observed in RBC aggregation - its decrease was significantly greater in old RBCs. It is important to note that such microrheological responses of different age subpopulations of cells were observed both to GT donors and stimulators of their endogenous synthesis.
Conclusion: Taken together the data obtained in the study indicate that old red blood cells were more sensitive to the action of donors of three gasotransmitters and stimulants of their endogenous synthesis. Under the influence of GTs, the increase in the deformability of old RBCs was such that it exceeded the value of this characteristic of the general population of mature RBCs.
