Effect of gasotransmitters (NO and H2S) on the red blood cell micromechanical properties in normal conditions and with type 2 diabetes mellitus

Резюме: Disorders of the rheological properties of blood in type 2 diabetes mellitus (DM-2T) can lead to a decrease in microvascular perfusion. There are publications that report on the effect of gasotransmitters (NO and H2S) on red blood cell (RBC) microrheology. However, the metabolism of these signaling molecules is impaired in diabetes. The purpose of this study was to investigate RBC microrheological responses to gasotransmitter donors of healthy individuals and DM-2T patients. RBC microrheology was recorded after cell incubation with: 1) NO donor – sodium nitroprusside (SNP, 100 M); 2) hydrogen sulphide donor – sodium hydrosulfide (NaHS, 100 M); 3) methylene blue, as a soluble guanylate cyclase (s-GC) inhibition (50 M); 4) glibenclamide, as K+ATP channel blocker (50 M). Cells were incubated for 30 min at 37°С. RBC suspension prepared in drug-free solution was used as a control sample.
Significant negative changes in the parameters of the hemorheological profile in patients with diabetes, including erythrocyte microrheology, were found. It was shown that an exposure of RBCs to both types of the gasotransmitter (GT) donors led to significant positive changes in the RBC microrheological properties. However, in diabetics, these changes were less pronounced. The analysis of the data obtained showed that the molecular intracellular targets for the action of GT can be either s-GC, ATP sensitive K+ channels, or elements of signaling pathways associated with the cellular membrane. The latter was indicated by microrheological responses of recovered red blood cell ghosts. Taken together the data obtained in this study indicate a statistically significant effect of donors of two gas transmitters on the micromechanical properties of erythrocytes. At the same time, in conditions of diabetes mellitus of the second type, these regulatory possibilities for the fluidity of whole blood and its transport potential are reduced.
